the E5-4XXX are meant to be in quad cpu systems, but will they work with Supermicro dual cpu motherboards ?

That's an "up to" spec. You can use these CPUs in single- dual- and quad-socket configurations.
The other way around is restricted, i.e. you can't use a Xeon E5-2xxx CPU in quad-socket configurations.
